OncoBayes Advances OB-001 to Address Oncology Challenges
OncoBayes, a clinical-stage pharmaceutical company, has completed preclinical studies for its product OB-001. This advancement targets two major unmet needs in oncology: resistance to Antibody-Drug Conjugates (ADC) and brain penetration of tyrosine kinase inhibitors (TKIs). OB-001 works as a selective inhibitor of P-gp and BCRP efflux pumps found in both blood-brain barriers and tumor cells.
In mouse studies, OB-001 increased brain exposure to kinase inhibitors, notably boosting osimertinib's brain exposure by nearly 400%. Concurrently, systemic exposure remained stable, potentially enhancing progression-free survival in EGFRm NSCLC patients. Additionally, OB-001 curtails efflux of ADC payloads like SN-38, DM4, and DXd, addressing ADC resistance mechanisms.
Future clinical trials will focus on proof-of-concept data with osimertinib and evaluate T-DXd re-sensitisation. OncoBayes aims for OB-001 to become a first-in-class oral adjunct in oncology therapies.
